This document is applicable to the quarantine detection and identification of grape bitter rot pathogens in tissues such as leaves, stems and fruits of host plants.

Chinese name. Grape bitter rot.
Scientific name. Greeneriauvicola (Berk.
Classification. Fungi, Ascomycota, Pezizomycotina, Sordar-
iomycetes), Diaporthales, Melanconielaceae, Greeneria.

5 Identification Principles
The symptoms of grape bitter rot pathogens in harming the host, the morphological characteristics of the pathogen and the molecular biological characteristics are the basis for identifying the pathogen.
6 Instruments and equipment
6.1 Biological microscope. with measurement function.
6.2 Dissecting microscope. minimum 10×.
6.3 Constant temperature and light incubator. 5℃~60℃, temperature accuracy ±0.1℃.
6.4 Electronic balance. sensitivity 0.001g.
6.5 Autoclave. Maximum pressure is 0.235MPa, and the adjustable temperature range is 105℃~135℃.
6.6 Desktop centrifuge. speed range is between 10000r/min and 30000r/min.
6.7 pH meter. range. pH 0.00~14.00, accuracy. ±0.01pH.
6.8 Constant temperature water bath. temperature range 4℃~100℃, temperature control accuracy ±0.3℃.
6.9 PCR instrument. temperature range 4℃~100℃, temperature uniformity ±0.4℃.
