1 Scope
This document describes a method for the determination of the count of Candida utilis in microbial preparations for feed.
This document applies to the determination of feed microbial preparations containing Candida utilis.

3 Terms and definitions
There are no terms or definitions that require definition in this document.
4 Reagents or materials
Unless otherwise specified, only analytical grade reagents were used.
4.1 Water. GB/T 6682, grade 3.
4.2 0.85% sterile saline. Accurately weigh 8.5 g of sodium chloride and add it to 1000 mL of water. Stir until completely dissolved. After packaging, place the mixture at 121 °C.
Sterilize for 30 min and set aside.
4.3 Red Bengal agar medium. see Appendix A, A.1.
4.4 Yeast peptone dextrose (YPD) agar medium. see A.2.
5.Instruments and equipment
In addition to routine sterilization and culture equipment in microbiology laboratories, other instruments and equipment are as follows.
5.1 Incubator. controllable accuracy ±1 ℃.
5.2 Balance. sensitivity is 0.1 g.
5.3 Optical microscope. 400x to 1000x.
5.4 Slap-type homogenizer and sterile homogenizing bag.
5.5 Vortex mixer.
5.6 Sterile culture dish. 90 mm in diameter.
5.7 Polymerase chain reaction (PCR) instrument.
6 Sampling
Implement in accordance with GB/T 42959.
