1 Scope
This document describes a HPLC method for the determination of flavonol glycosides in the feed additive Epimedium brevicornum extract.
This document applies to the five flavonol glycosides (icariin, epimedin A, epimedin B, epimedin C) in the feed additive epimedin extract.
and Baohuoside I).
In this document, the limits of quantification for icariin, epimedin A, epimedin B, epimedin C and baucariside I are all 0.2 g/kg.

3 Terms and definitions

4 Principle
The flavonol glycosides in the samples were extracted with methanol solution and determined by high performance liquid chromatography using the external standard method.
5 Reagents or materials
Unless otherwise specified, only analytical grade reagents were used.
5.1 Water. GB/T 6682, Grade 1.
5.2 Methanol. chromatographic grade.
5.3 Acetonitrile. chromatographic grade.
5.4 70% methanol solution. Measure 700 mL of methanol (5.2) into a 1000 mL volumetric flask, add water to make up to volume, and mix well.
5.5 Standard stock solution (2 mg/mL). Accurately weigh icariin (CAS. 489-32-7, purity ≥ 96%), epimedin A (CAS.
110623-72-8, purity ≥95%), epimedin B (CAS. 110623-73-9, purity ≥95%), epimedin C (CAS. 110642-44-9, purity
20 mg each (accurate to 0.01 mg) of baohuoside I (CAS. 113558-15-9, purity ≥ 95%) were placed in 10 mL brown
Dissolve in methanol (5.2) in a volumetric flask and make up to volume, mix well. Store below -18℃, valid for 3 months.
5.6 Mixed standard intermediate solution (200 mg/L). Accurately pipette 1 mL of each standard stock solution (5.5) into a 10 mL brown volumetric flask.
Add 70% methanol solution (5.4) to the volume and mix well. Store at 2℃~10℃, valid for 3 months.
5.7 Mixed standard series solution. Accurately pipette an appropriate amount of mixed standard intermediate solution (5.6) and prepare it with 70% methanol solution (5.4) to a mass concentration of
