4 Principle
Bee venom hemolytic peptide is the main biologically active substance of bee venom. The bee venom hemolytic peptide in the sample is easily soluble in water, and the maximum ultraviolet absorption wavelength is
220nm. Therefore, the bee venom dry powder sample is dissolved in pure water, and the extract is filtered and measured by a high-performance liquid chromatograph equipped with an ultraviolet detector.
Quantification by external standard method.
5 Reagents and materials
The water is the first grade water that meets the requirements of GB/T 6682.
5.1 Reagents
5.1.1 Acetonitrile. chromatographically pure.
5.1.2 Trifluoroacetic acid. chromatographically pure.
5.2 Standard products
Bee venom hemolytic peptide (melittin, CAS. 20449-79-0) purity ≥ 97%.
5.3 Preparation of solution
5.3.1 Bee venom hemolytic peptide standard stock solution (10 mg/mL). accurately weigh 100 mg of the standard substance of bee venom hemolytic peptide into a 10 mL volumetric flask
