α-amanitin, etc. in forensic science biological examination materials
Detection of 5 Amanita Toxins by Liquid Chromatography-Mass Spectrometry
1 Scope
This document stipulates α-amanitin, β-amanitin, and γ-amanitin in biological test materials (blood, urine, liver, etc.) in the field of forensic science.
Liquid chromatography-mass spectrometry (LC-MS) detection method for five kinds of amanitaxins including peptide, carboxydihydroxyphallulin, and dihydroxyphallulin.
This document is applicable to α-amanitin, β-amanitin, and γ-amanitin in biological test materials (blood, urine, liver, etc.) in the field of forensic science.
Qualitative and quantitative analysis of five kinds of amanitaxins including peptide, carboxydihydroxyphallulin, and dihydroxyphallulin. Other inspection materials can refer to
use.

3 Terms and definitions
The terms and definitions defined in GA/T 122 apply to this document.
4 Principles
Using blank samples and added samples as controls, according to the requirements of parallel operations, five kinds of Amanita toxins were tested on biological test materials (blood, urine, liver, etc.)
Extract, purify and concentrate compounds (see Appendix A for basic compound information). Liquid chromatography-mass spectrometry was used for detection, with retention time and mass spectrometry characteristics
The ion pair and ion pair abundance ratio are used as the basis for qualitative judgment; the chromatographic peak area is used as the basis, and the external standard single-point calibration method is used for quantification.
analyze.
5 Reagents and materials
5.1 Reagents
The experimental water should comply with the first-class water specified in GB/T 6682.Unless otherwise stated, the reagents used in the analysis are of chromatographic grade.
Reagents include.
a) Acetonitrile;
b) 10% methanol solution (take the preparation of 100mL as an example). measure 10mL methanol and 90mL water, and mix;
c) 1% formic acid acetonitrile solution (take preparation of 100mL as an example). measure 1mL formic acid and 99mL acetonitrile and mix evenly;
d) 5% methanol solution (take preparation of 100mL as an example). Measure 5mL methanol and 95mL water and mix well;
