4 principles
Using the blank sample and the added sample as a control, according to the requirements of parallel operation, 9 kinds of barbiturates (compounds) in biological samples (blood) were tested:
For the basic information of the substance, see Appendix A) for extraction, purification and concentration: Detection by liquid chromatography-mass spectrometry, retention time, mass spectrometric characteristic ions
The abundance ratio of pair and ion pair was used as the basis for qualitative judgment; the chromatographic peak area ratio was used as the basis for quantification, and internal standard-single-point calibration method or internal standard-calibration
Quantitative analysis was performed by the quasi-curve method:
5 Reagents and Materials
5:1 Reagents
The experimental water should meet the first-class water specified in GB/T 6682: Unless otherwise stated, the reagents used in the analysis were of analytical grade:
Reagents include:
a) Methanol;
b) Acetonitrile: chromatographically pure;
c) Extraction solvent (take the preparation of 100mL as an example): measure 90mL of ethyl acetate and 10mL of n-hexane, and mix well;
d) 0:1mol/L hydrochloric acid solution (take the preparation of 100mL as an example): pipette 0:82mL concentrated hydrochloric acid (analytical pure, mass fraction 36%~
38%), dilute with water and dilute to 100mL, mix well;
