1 Scope
This standard specifies the method for the determination and analysis of the proportion of nicotine optical isomers of tobacco characteristic components.
This standard applies to tobacco and tobacco products, electronic cigarette smoke, smokeless tobacco products and other samples of tobacco characteristic components nicotine rotation
For the determination of the proportion of the structure, the gum base type tobacco product can be used for reference. Among them, high performance liquid chromatography is suitable for nicotine optical isomer ratio in samples.
For the routine detection of the example, ultra-high performance convergent-phase chromatography-tandem mass spectrometry is suitable for the confirmatory detection of the proportion of nicotine optical isomers in the sample.

3 Principle
The nicotine in the sample is extracted with a mixed solution of n-hexane and methanol under an alkaline solution, and the extract is subjected to matrix-dispersed solid phase extraction.
After normalization, the positive-phase liquid chromatography UV detector was used to detect the (S)-(-)-nicotine and tobacco in tobacco and tobacco products by external standard method.
(R)-()-nicotine content, then calculate the proportion of nicotine optical isomers.
4 reagents
Analytical grade or higher reagents should be used unless otherwise specified.
4.1 Water, GB/T 6682, level one.
4.2 Methanol.
4.3 n-hexane.
4.4 Sodium hydroxide.
4.5 5% aqueous sodium hydroxide, weigh 5.0g of sodium hydroxide (4.4), completely dissolved with water (4.1), transferred to 100mL plastic capacity
In the bottle, dilute to the mark with water (4.1).
4.6 n-hexane and methanol solution (volume ratio of 98.2), pipet 20mL of methanol (4.2) into a 1000mL volumetric flask, with n-hexane
(4.3) Make up to the scale.
4.7 Graphitized carbon black (GCB).
4.8 N-propyl ethylenediamine bonded silica gel (PSA).
4.9 Anhydrous magnesium sulfate (MgSO4).
4.10 (S)-(-)-nicotine standard, the purity is not less than 99%.
4.11 (R)-()-nicotine standard, the purity is not less than 99%.
