1 Scope
This standard specifies a high performance liquid chromatographic method food exports Acid Orange Ⅱ number.
This standard is applicable to the detection of drinks, wine, chili sauce, fish and braised foods Acid Orange Ⅱ number.
Principle 2
The sample was ethanol - ammonia solution was extracted with an amino anion exchange solid-phase extraction (SPE), reversed-phase high performance liquid chromatography - diode array detection
Chromatographic analysis is performed using external standard.
3 Reagents and materials
Unless otherwise specified, use only analytical reagent.
3.1 Methanol. chromatographically pure.
3.22% formic acid. Take 2mL formic acid, water volume to 100mL.
3.3 10% methanol aqueous ammonia solution. Take 10mL ammonia, methanol and dilute to 100mL.
3.4 ethanol - ammonia solution. Take 20mL ammonia was added 70mL of ethanol, water volume to 100mL, shake reserve.
3.5 0.02mol/L ammonium acetate solution. Weigh accurately 1.54g ammonium acetate, add water to 1000mL, dissolved by 0.45μm membrane filter.
3.6 amino anion exchange solid phase extraction column. 500mg, 3mL, or equivalent person. Before using successively with 3mL methanol, 3mL2% formic acid
Aqueous activation.
3.7 Acid Orange Ⅱ on standard stock solution. Weigh accurately Acid Orange Ⅱ number 10.0mg (purity ≥95%), dissolved in water volume to 100mL, dubbed
100μg/mL standard stock solution.
3.8 Acid Orange Ⅱ number of standard working solution. Pipette appropriate amount of standard stock solution successively diluted with water to a concentration of 0.1μg/mL, 0.2μg/mL,
0.5μg/mL, 1.0μg/mL, working standard solution 10.0μg/mL, and stored at 4 ℃ refrigerator.
