Paratuberculosis bacteriological examination operation procedures
1 Scope
This standard specifies the technical regulations for the operation of bacteriological examination of paratuberculosis.
This standard applies to the detection of tuberculosis in cattle, sheep and deer.
2 reagents and materials
2.1 Reagents
2.1.1 75% alcohol swabs.
2.1.2 Lipase.
2.1.3 0.5% Tween-80 in PBS (0.5 mL Tween-80 dissolved in 100 mL of PBS pH 7.0).
2.1.4 Diethyl ether-ethanol aliquot (diethyl ether 20 mL, ethanol 20 mL mixed).
2.1.5 Oxalic acid, malachite green mixture (weigh 10g of oxalic acid, 0.02g of malachite green dissolved in 100mL of distilled water, filtered with a pore size of 0.45μm
Membrane filtration sterilization).
2.1.6 amphotericin B, neomycin mixture (weigh amphotericin B5mg, neomycin 5mg dissolved in 100mL distilled water, with pore size

3 sample collection and processing
3.1 Sample collection
3.1.1 Latex
The normal milk is collected within 14 days to 25 days after delivery. When taking the milk sample, the nipple is first scrubbed and disinfected 3 times to 4 times with a 75% alcohol cotton swab.
After the first few colostrums, approximately.200 mL of the milk sample was collected in a sterile container. Sampling should be completed within 15 minutes.
3.1.2 Feces
Choose feces mixed with mucus, blood, mucous membranes or use a curette to take a small amount of mucus feces from the deep rectum (about 30cm) of the animal.
In the container; the above materials can also be directly made into several smears, naturally dried, and wrapped with waterproof paper for inspection.
3.1.3 Intestines and lymph nodes
Intestinal segments with obvious lesions, ileocecal valves, and lymph nodes adjacent to the intestine are selected, and the feces are rinsed from the intestines with tap water.
