Foreword
This standard was drafted in accordance with the rules given in GB/T 1.1-2009.
This standard replaces SN/T 1172-2003 "Agar leukemia detection method agar immunodiffusion test".
Compared with SN/T 1172-2003, the main technical changes of this standard are as follows.
--- Increased isolation and identification of chicken leukemia virus;
--- Increased polymerase chain reaction;
--- Increased fluorescence quantitative polymerase chain reaction;
--- Increased enzyme-linked immunosorbent assay.

1 Scope
This standard specifies the isolation and identification of chicken leukemia virus, polymerase chain reaction, fluorescence quantitative polymerase chain reaction, enzyme-linked immunosorbent assay
Adsorption test, agar diffusion test detection technology.
This standard applies to the quarantine of chicken leukemia.

4 Virus isolation and identification
4.1 Reagents and materials
4.1.1 Main reagents. DMEM cell culture medium, 0.25% trypsin, penicillin (100000 IU/mL), streptomycin (100000 IU/mL),
Fetal bovine serum, cell lysate.
4.1.2 Main equipment and equipment. cell culture incubator, inverted microscope, microplate reader, centrifuge, grinder, cell culture flask.
4.1.3 Cells and cell lines. chicken embryo fibroblasts (see Appendix A), chicken embryo fibroblasts spontaneous immortal (DF1 cell line).
4.2 Collection, storage and transportation of samples
Mainly collect chicken whole blood, tissue samples (spleen, liver and kidney) or throat swabs, cloaca swabs. Sample storage and shipping

4.3 Sample processing
4.3.1 Whole blood. Collect whole blood samples, put them into the test tube with anticoagulant, mix gently; place the test tube upright at room temperature or 37 °C incubator
