1 Scope
This standard specifies Cereals by amitrole residues of liquid chromatography and quantitative detection of liquid chromatography tandem mass spectrometry qualitative and quantitative detection

3 The first method by high performance liquid chromatography (HPLC method)
3.1 Principle
Samples amitrole methylene chloride, a mixed solution of 1% acetic acid extraction, mixed cation exchange column purified and concentrated by fluorescence amine derivative
Health, acetonitrile and phosphate buffer as the mobile phase was determined by HPLC with fluorescence detector, external standard.
3.2 Reagents and materials
Unless otherwise specified, all reagents of proven analytical grade water should comply with GB/T 6682's.
3.2.1 acetonitrile. chromatography.
3.2.2 dichloromethane. chromatography.
3.2.3 acetone. chromatography.
3.2.4 n-hexane.
3.2.5 glacial acetic acid.
3.2.6 acetate buffer. sodium acetate solution 0.05mol/L in acetic acid solution 800mL0.05mol/L in to
1000mL.
3.2.7 Phosphate buffer. 10% phosphoric acid was added 0.05mol/L sodium phosphate solution adjusted to pH 3.0.
3.2.8 basic methanol. ammonia in methanol (595, volume ratio).
3.2.9 PCX mixed cation exchange column.
3.2.10 fluorescamine. greater than 98% purity.
3.2.11 amitrole standards. purity of greater than 97%.
3.2.12 standard stock solution. Weigh amitrole standards 10.0mg, methanol dubbed 0.1mg/mL standard stock solution.
3.2.13 standard working solution. According to the standard stock solution need to take the appropriate volume, with the working standard solution is made to match the flow of the working fluid active
