Canine infectious hepatitis diagnosis technology
1 Scope
This standard specifies the clinical diagnosis of canine infectious hepatitis (ICH), isolation and identification of canine infectious hepatitis virus (ICHV), and enzyme-linked immunoassay.
Diagnostic techniques such as ELISA and immunohistochemistry.
This standard applies to the diagnosis of canine infectious hepatitis.
2 clinical diagnosis points
2.1 Clinical symptoms

3 Virus isolation and identification
3.1 Material preparation
3.1.1 Reagents

3.1.1.2 Canine kidney passage cells (MDCK cells).
3.1.1.3 Standard positive serum. ICHV monoclonal antibody, neutralizing antibody titer 1.1024.
3.1.1.4 Standard negative serum. Canine serum without ICHV infection.
3.1.1.5 newborn bovine serum, penicillin, streptomycin.
3.1.2 Equipment
a) cell culture flask;
b) a straw;
c) a carbon dioxide incubator;
d) 37 ° C constant temperature water bath;
e) ordinary refrigerators and low temperature refrigerators;
f) centrifuges and centrifuge tubes;
g) grinding equipment;
h) ordinary optical microscope;
i) micro-sampler, capacity 5μL ~ 50μL, 50μL ~.200μL;
j) 0.45 μm microporous membrane.
3.1.3 Sample
ICHV is present in tissues and organs such as tonsil, liver and spleen of sick dogs. Sterile collection of these organs made with serum-free DMEM
