Technical specification for identification of apricot varieties
SSR molecular marker
1 Scope
This standard specifies the use of simple repeat sequences (SSR) molecular markers to identify apricots
(Armeniacacathayana D.L. Fuetal.) technical specifications of the variety.
This standard applies to the identification of SSR molecular fingerprints of apricot varieties.
2 Terms and definitions
The following terms and definitions apply to this document.
2.1
Test species testcultivar
The variety of apricots to be identified shall be provided by the examiner.
2.2
Control variety comparingcultivar
For the varieties that are used for fingerprint comparison with the varieties to be inspected (the varieties to be inspected are new varieties, the control varieties are similar varieties; the varieties to be inspected
For counterfeit varieties, the control variety is a true variety), which is provided by the examiner or others.
2.3
Core primer coreprimer
The amplified product has high stability and consistency (no interference band) and has strong discriminating ability, which can be used to establish apricot
Synthetic primers for fingerprint alignment.
2.4
Fingerprint SSRfingerprint
DNA fragments of the tested and control varieties amplified by SSR core primers (or equivalent to core primers), passed through polypropylene
Amide electrophoresis or capillary electrophoresis to obtain maps of DNA fragments of different sizes.
2.5
Fingerprint comparison SSRfingerprinting
Comparing the SSR fingerprints of the tested varieties with the control varieties, and analyzing whether the SSR fingerprints of the tested and control varieties have
There are differences and their size is determined.
3 Principle
Simple repeat sequence molecular markers (SSRs), often referred to as "microsatellite markers," are distributed at different locations throughout the genome of apricot kernels.
The number of repeating units at each locus of different varieties may be different, thus forming a polymorphism of the SSR marker. Due to each simple repeat
The sequences at both ends are highly conserved single-copy sequences, so specific primers can be designed based on the sequences at both ends, using PCR to
The DNA repeats between the primers are amplified, and the amplified product fragments of different sizes are separated by electrophoresis and labeled with fluorescent dyes.
distinguish. Therefore, according to the polymorphism of the SSR locus, the apricot variety can be identified by PCR amplification and electrophoresis.
