3 Terms and Definitions
The following terms and definitions apply to this document.
3.1
glucose oxidase
A dehydrogenase that specifically catalyzes the conversion of β-D-glucose to D-gluconic acid and hydrogen peroxide in the presence of oxygen.
3.2
At pH 5.1 and 35°C, 1.0 μmol of β-D-glucose can be oxidized into D-gluconic acid and hydrogen peroxide per minute.
The amount of enzyme required is one activity unit (U).
4 Principles
Under the action of glucose oxidase, glucose reacts with oxygen to produce gluconic acid and hydrogen peroxide, hydrogen peroxide and colorless reduced form.
Under the action of peroxidase, o-dianisidine generates water and red oxidized o-dianisidine, which is a red substance at a wavelength of 500nm.
There is a specific absorption peak, and the number of activity units of the enzyme can be calculated by measuring the rate of increase of the absorbance of the red substance at 500nm.
5 Reagents or materials
5.1 Water
Grade II water in accordance with GB/T 6682.
5.2 50mmol/L sodium acetate buffer
Weigh 0.82g of anhydrous sodium acetate, dissolve it in 150mL of pure water, adjust the pH to 5.1 with 1mol/L hydrochloric acid at 35℃, and dilute to 5.5ml.
