4 Principle
After the sample was saponified, the astaxanthin esters were converted into free astaxanthin.
The ionization was carried out, the detection was carried out by diode array detector, the qualitative analysis was carried out by retention time and chromatogram, and the quantitative analysis was carried out by internal standard method.
5 Reagents and Materials
5.1 Water. GB/T 6682, Grade 1.
5.2 Astaxanthin standard substance. purity not less than 95%, CAS number 472-61-7.
5.3 β-Apocarotene aldehyde standard substance. purity not less than 95%, CAS number is 1107-26-2.
5.4 Acetone. high purity.
5.5 Methanol. chromatographic grade.
5.6 Dichloromethane. high purity.
5.7 Sodium hydroxide. analytical grade.
5.8 Butylated hydroxytoluene (BHT). analytical grade.
5.9 Phosphoric acid. analytical grade.
5.10 Methanol-dichloromethane solution. Take 750 mL of methanol and 250 mL of dichloromethane, add 0.2 g of BHT, and mix well.
