Microbial source antibiotic secondary metabolite
Nematicidal activity assay
1 Scope
This standard specifies the method for determining the nematicidal activity of microbial-derived antibiotic secondary metabolites by the dip method.
This standard is applicable to the determination of nematicidal activity of secondary metabolites of microbial source antibiotics.

3 Terms and definitions
The following terms and definitions apply to this document.
3.1
Microbial source antibiotic secondary metabolite microbialantibioticsecondarymetabolites
A substance produced by secondary metabolism of microorganisms having antibiotic activity characteristics.
3.2
Lethal concentration of lethalconcentration 50%; LC50
The concentration of microbial-derived antibiotic secondary metabolites that caused half of the nematodes to die.
4 Principle
Insect nematodes are immersed in microbial-derived antibiotic secondary metabolites to cause nematodes to die, and nematode dead cells or tissues are stained with neutral red
Red color visible under the microscope, judging the lethal condition of secondary metabolites to nematodes by dyeing, calculating the corrected death of nematodes
Rate, according to the linear regression relationship between the corrected mortality rate and the logarithm of the drug concentration, the regression curve is obtained, and the LC50 is calculated to determine
Microbial source antibiotic secondary metabolite nematicidal activity.
5 reagents or materials
Analytically pure reagents are used unless otherwise specified.
5.1 Water. GB/T 6682, secondary water.
5.2 5mg/mL cholesterol solution.
Weigh the appropriate amount of cholesterol, dissolve in the corresponding volume of absolute ethanol, and prepare a 5mg/mL cholesterol solution, 0.22μm pore size micropores
The filter membrane is sterilized by filtration.
5.3 1mol/L calcium chloride solution.
Weigh the appropriate amount of calcium chloride, dissolve it in the corresponding volume of water, prepare a 1mol/L calcium chloride solution, filter with a 0.22μm pore size filter.
