1 Scope
This document describes a method for the determination of puerarin in health foods by HPLC.
This document applies to puerarin in health foods in dosage forms such as soft capsules, hard capsules, tablets, powders, granules, oral liquids, and alcoholic preparations.

4 Principle
Puerarin in the sample was extracted with 50% methanol solution, purified by solid phase extraction column if necessary, separated by high performance liquid chromatography, and detected by ultraviolet light.
The samples were detected by the external standard method, and the qualitative analysis was based on the retention time.
5 Reagents and Materials
Unless otherwise specified, only analytical grade reagents were used.
5.1 Reagents
5.1.1 Water. Grade 1 water as specified in GB/T 6682.
5.1.2 Methanol (CH4O). chromatographically pure.
5.1.3 Methanol (CH4O).
5.1.4 Acetonitrile (CH3CN). chromatographic grade.
5.1.5 Phosphoric acid (H3PO4). high purity.
5.1.6 Petroleum ether. boiling range 30℃~60℃.
5.2 Reagent preparation
5.2.1 0.1% (volume fraction) phosphoric acid solution. measure 1.0 mL of phosphoric acid (5.1.5), dilute to 1000 mL with water, and mix well.
5.2.2 50% (volume fraction) methanol solution. Take 50 mL of methanol (5.1.3), dilute to 100 mL with water, and mix well.
