1 Scope

This document describes the HPLC and spectrophotometric determination of D-biotin in vitamin premixes and compound premixes. method. This document is applicable to the determination of D-biotin in vitamin premix feeds and compound premix feeds. The detection limit of the HPLC method in this document is 1 mg/kg, and the quantification limit is 3 mg/kg; the detection limit of the spectrophotometric method is 1.5 mg/kg, limit of quantification is 5 mg/kg.

There are no terms or definitions that require definition in this document. 4 High-performance liquid chromatography 4.1 Principle The D-biotin in the sample was extracted with water and determined by high performance liquid chromatography using the external standard method. 4.2 Reagents or materials Unless otherwise specified, only analytical grade reagents were used. 4.2.1 Water. GB/T 6682, Grade 1. 4.2.2 Acetonitrile. chromatographic grade. 4.2.3 Diethylenetriaminepentaacetic acid (DTPA). 4.2.4 5 mol/L sodium hydroxide solution. Weigh 20 g of sodium hydroxide, add appropriate amount of water to dissolve, cool to room temperature, and dilute to 100 mL. 4.2.5 0.05% trifluoroacetic acid solution. Measure 0.5 mL of trifluoroacetic acid, dilute to 1000 mL with water, and adjust with sodium hydroxide solution (4.2.4). Adjust pH to 2.5. 4.2.6 Standard stock solution (0.2 mg/mL). Weigh 10 mg of D-biotin (CAS No. 58-85-5, purity not less than 98%) standard (accurate to 0.01 mg) in a 50 mL volumetric flask, add water, ultrasonically dissolve, dilute to volume with water, and mix well. Store at 2°C~8°C. 3 months. 4.2.7 Standard intermediate solution (20 μg/mL). Accurately pipette 5 mL of D-biotin standard stock solution (4.2.6) into a 50 mL volumetric flask. Dilute to volume with water. Store at 2°C~8°C. Valid for 2 months.
