Detection method
1 Scope
The first method of this standard stipulates the liquid chromatography-tandem mass spectrometry method for the determination of bacitracin A in the blood and feed of imported and exported food animals, and the second method.
The law stipulates an ELISA method for the detection of bacitracin in food animals and feeds.
This standard applies to the determination of bacitracin in the blood and feed of imported and exported food animals (pig, cattle, sheep, poultry).
This standard applies to entry and exit inspection and quarantine work.

3 method summary
After the sample is pulverized and homogenized, the residue of bacitracin A in the sample is extracted with a methanol-formic acid aqueous solution, and purified by a solid phase extraction cartridge.

4 reagents
Unless otherwise stated, the reagents used were of analytical grade and the water was the first grade water specified in GB/T 6682.
4.1 Dichloroacetic acid.
4.2 EDTA.
4.3 TCA.
4.4 Methanol. Chromatographically pure.
4.5 Acetonitrile. Chromatographically pure.
4.6 Formic acid. chromatographically pure.
4.7 1.5mmol/LEDTA solution. Weigh 0.5g EDTA into a volumetric flask and dilute to 1000mL with water.
4.8 1% TCA solution. Weigh 10.0g of TCA in a volumetric flask and dilute to 1000mL with water.
4.9 0.1% formic acid-water solution. Pipette 1 mL of formic acid into a volumetric flask and dilute to 1000 mL with water.
4.10 4% trichloroacetic acid-acetonitrile solution. Pipette 4 mL of dichloroacetic acid in a volumetric flask and make up to 100 mL of acetonitrile.
4.11 Constant volume solution. 0.1% formic acid-water solution acetonitrile volume ratio of 8.2.
4.12 Bacitracin A standard. purity ≥ 96.0%.
4.13 Standard stock solution. Accurately weigh the bacitracin standard, dissolve to a volume of 10 mL with methanol (4.3), and the solution concentration is 1000 μg/mL.
