Acid hydrolysis method for impurity determination
1 Scope
This document describes a test method for determining animal-derived impurities in floury grains using acid hydrolysis.
This document applies to the determination of lipophilic animal-derived impurities such as insect fragments and rodent hair in wheat flour and rice flour.
The determination of lipophilic animal-derived impurities in white powdery grains was performed in accordance with the prescribed method.

3 Terms and Definitions

4.Principles
Starch and protein are removed from floury grains by acid hydrolysis. Animal-derived impurities are extracted from the aqueous phase using the oil phase. The oil phase is then filtered.
After collecting impurities with filter paper, observe and record them under a microscope.
5.Reagents and Materials
Unless otherwise specified, all reagents used in this method are of analytical grade, and the water should meet the requirements of Grade III water as specified in GB/T 6682.
5.1 Reagents
5.1.1 n-Heptane (C7H16).
5.1.2 Liquid paraffin. relative density 0.83~0.89.
5.1.3 Hydrochloric acid (HCl).
5.1.4 Sodium dodecyl sulfate (C12H25SO4Na).
5.1.5 Glycerol (C3H8O3).
5.1.6 Anhydrous ethanol (C2H6O).
5.2 Reagent Preparation
5.2.1 Hydrochloric acid solution (5%). Take 5 mL of hydrochloric acid (5.1.3) and add water to make up to 100 mL.
