1 Scope
This document specifies the principles, reagents and methods for the determination of lycopene, lutein and carotene in tomato products by ultra-high performance liquid chromatography.
Materials, equipment, analytical procedures, calculation of results, precision, limits of detection, and limits of quantification.
This document applies to lycopene, lutein, alpha-carotene and beta-carotene in tomato powder, tomato paste, tomato beverages, and dried tomato products

4 Principles
After adding water and ethanol to the sample, extract the lycopene, lutein and carotene in it with ether-n-hexane-cyclohexane mixed solution.
Separation by high performance liquid chromatography, detection by UV detector or diode array detector, and quantification by external standard method.
5 Reagents and Materials
Unless otherwise specified, use only chromatographically pure reagents.
5.1 Water. GB/T 6682, Class I.
5.2 Cyclohexane (C6H12).
5.3 Diethyl ether [(C2H5)2O].
5.4 n-hexane (C6H14).
5.5 Anhydrous ethanol (C2H5OH). analytically pure.
5.6 Dibutylhydroxytoluene (C15H24O, BHT). analytically pure.
5.7 Methanol (CH3OH).
5.8 Dichloromethane (CH2Cl2).
