Determination of soy sauce sodium iron EDTA iron-fortified
1 Scope
This standard specifies the method for determination of iron fortified soy sauce sodium iron EDTA (NaFeEDTA) of.
This standard applies to the determination of iron fortified soy sauce sodium iron EDTA.
This standard is the first detection limit was 0.030μg/mL, when sampling is 2.5mL, the minimum detectable concentration of 6.0μg/mL, the best
The linear range of 0.2μg/mL ~ 20μg/mL. The minimum detection limit of the second method is 0.24μg/mL, when sampling is 3.0mL, the lowest
Detectable concentration of 40μg/mL, the optimal linear range of 4.0μg/mL ~ 24μg/mL.
The first method HPLC
Principle 2
Iron fortified soy sauce sample, after precipitated in methanol, filtered, diluted with ion-pair reversed-phase high performance liquid chromatography in sodium iron EDTA
The FeEDTA- and separation of impurities in the sample, the UV detector at 254nm wavelength detection, based on peak retention time qualitative criteria
Curve peak area.
3 Reagents
The reagents used in the process, unless otherwise specified, reagents were of analytical grade, distilled water or water of equivalent purity water.
3.1 Methanol. chromatographically pure.
3.2 40% tetrabutyl ammonium hydroxide solution (TBAOH).
Dilute acid solutions 3.3. The formulation was diluted 10-fold with water of 88% formic acid.
3.4 sodium iron EDTA standard. purity (to NaFeEDTA · 3H2O meter) ≥99.0%.
3.5 75% methanol solution. a volume of 750mL of methanol was added to a volume of 250mL of water, and mix.
3.6 sodium iron EDTA standard stock solution. accurately weighed sodium iron EDTA standard 0.1g, into 50mL brown container
Flask, dissolved in water and diluted to the mark, shake, solution concentration of 2000μg/mL. From light in the refrigerator, it is recommended 15 days
use.
3.7 EDTA iron sodium intermediate standard solution. Imbibe sodium iron EDTA standard stock solution 2.50mL, placed
50mL brown volumetric flask, with 75% methanol solution to volume, shake, and this solution at a concentration of 100μg/mL. From light in the refrigerator, it is recommended
15 days of use.
3.8 sodium iron EDTA solution using the standard. Pipette iron EDTA sodium intermediate standard solution, 1.0mL, 2.0mL,
4.0mL, 6.0mL, 8.0mL, 10.0mL in 50mL volumetric flask, add water to volume, after shaking, to obtain EDTA
Sodium iron standard series, each containing NaFeEDTA · 3H2O was 2.0μg/mL, 4.0μg/mL, 8.0μg/mL, 12.0μg/mL,
