1 Scope

This section specifies the method of ELISA for melamine in dairy products. This section applies to the quantification of melamine in liquid milk and milk powder using ELISA.

3 Principle

Coupling antigens are pre-coated in the microtiter wells of the microtiter plate, and residual melamine in the sample competes with the pre-coated conjugation antigen in micropores for cyanuric acid. Amine antibody, enzyme-conjugated secondary antibody, was developed with TMB substrate and the absorbance of the sample was negatively correlated with the content of melamine in the residue. The melamine content of the sample can be determined by multiplying the line by the corresponding dilution factor.

4 Reagents and materials

4.1 Melamine Enzyme Immunoassay Kit It is transported and stored at 2°C~8°C. The product is valid for 12 months. The kit reference SN/T 2775 was evaluated by the CNCA Commercial Evaluation Kit Expert Committee. See Appendix A for results. The kit consists of the following. a) melamine coated microtiter plates; b) melamine standard working fluid; c) melamine antibody working solution; d) enzyme-labeled secondary antibody working fluid; e) concentrated sample diluent; f) Concentrate the washing solution; g) substrate solution; h) Stop solution.
