1 Scope

This standard specifies the method for the determination of bacitracin zinc in feed by liquid chromatography. This standard applies to the determination of zinc content of bacitracin in pig and chicken additive premixes.

3 Principle of the method

The bacitracin zinc in the additive premix can be extracted with an acidified organic solvent system, centrifuged, and the supernatant is taken for liquid chromatography analysis.

4 reagents and materials

The following reagents are of analytical grade unless otherwise specified, and the water meets the requirements of GB/T 6682 secondary water. 4.1 Phosphate buffer. pH 6.0, 1.5 g of dipotassium hydrogen phosphate and 8.5 g of potassium dihydrogen phosphate were weighed and dissolved in 1 L of water. 4.2 Phosphate-EDTA buffer. pH 4.5, weigh 13.6g potassium dihydrogen phosphate and 2.5g disodium edetate dissolved in 1L In the water. 4.3 Bacitracin zinc standard solution. 4.3.1 Preparation of standard stock solution. Accurately weigh 150.0mg of bacitracin zinc standard, dissolve it with extraction solution, and make up to 100mL. The solution bacillus zinc concentration was 1500 mg/L. 4.3.2 Intermediate solution. Pipette 10 mL of standard stock solution (4.3.1) and dilute to 50 mL with extraction solution. The concentration of the solution bacitracin zinc is 300mg/L. 4.3.3 Working solution. Pipette the intermediate solution (4.3.2) 0.20mL, 1.0mL, 5.0mL, and dilute to 10mL of the solution bacillus with the extraction solution. The concentrations of the peptide zinc were 6 mg/L, 30 mg/L, and 150 mg/L, respectively. 4.4 Solvent system. Prepared according to Table 1. The volume of each solvent label is taken in a 100 mL volumetric flask and the volume is adjusted to the mark with water.
