1 Scope

This standard specifies the principle of post-column photochemical derivatization high performance liquid chromatography for the determination of aflatoxin content in food and edible vegetable oils. Reagents and materials, instruments and equipment, sample preparation, sample preparation, procedure, calculation of results and precision. This standard applies to the determination of aflatoxin content in grain, oil and products (except oil). The quantitative detection limits of aflatoxins B1, B2, G1 and G2 in this standard are 0.5μg/kg, 0.25μg/kg, 1.0μg/kg and 0.5 μg/kg.

3 Principle

The sample is extracted by methanol-water, and the extract is purified by filtration, dilution and immunoaffinity chromatography, and separated by high performance liquid chromatography. The photochemical derivatizer after the column is used to derivatize the isolate to generate a fluorescent signal, and the fluorescence intensity is measured by a fluorescence detector. The content of aflatoxin in the sample was calculated by the standard curve method.
