4 Principle

The sample was dissolved in 0.1% formic acid solution, and trigonelline was extracted and purified by cation solid phase extraction column. Quantitative determination by internal standard method.

5 Reagents and Materials

Unless otherwise specified, all reagents were of analytical grade and water was grade 1 water in accordance with GB/T 6682. 5.1 Reagents 5.1.1 Formic acid (CH2O2). chromatographically pure. 5.1.2 Methanol (CH3OH). chromatographically pure. 5.1.3 Ammonia water (NH3·H2O). 5.1.4 Acetonitrile (CH3CN). chromatographic grade. 5.1.5 Ammonium acetate (CH3COONH4). chromatographically pure. 5.2 Solution preparation 5.2.1 0.1% formic acid solution. Take 1 mL of formic acid (5.1.1) in a 1000 mL volumetric flask, dilute to volume with water, and mix well. 5.2.2 2% ammonia methanol solution. Take 2 mL of ammonia (5.1.3) in a 100 mL volumetric flask, add methanol (5.1.2) to the mark and mix. 5.2.3 80% methanol aqueous solution. Take 80 mL of methanol (5.1.2) in a 100 mL volumetric flask, dilute to the mark with water, and mix well. 5.2.4 5mmol/L ammonium acetate solution. weigh 0.385g of ammonium acetate (5.1.5) into a 1000mL volumetric flask, dissolve it in water and make up to 1/4 inch.
