1 Scope

This document specifies the levels of tryptamine, phenethylamine, putrescine, cadaverine, histamine, octopamine, tyramine, spermidine and spermine in feed ingredients of animal origin. High performance liquid chromatography method. This document is applicable to animal sources such as fish meal, shrimp meal, chicken meal, pork meal, meat and bone meal, blood meal, feather meal, fish melted pulp, squid paste and silkworm chrysalis meal Determination of nine biogenic amines including tryptamine, phenethylamine, putrescine, cadaverine, histamine, octopamine, tyramine, spermidine and spermine in sex feed ingredients. The limit of detection in this document is 5mg/kg, and the limit of quantification is 10mg/kg.

4 Principles

The biogenic amines in the samples were extracted with perchloric acid solution, derivatized with dansyl chloride, determined by high performance liquid chromatography, and quantified by external standard method.

5 Reagents or materials

Unless otherwise specified, only analytical grade reagents were used. 5.1 Water. GB/T 6682, Class I. 5.2 Acetonitrile. chromatographically pure. 5.3 Acetone. 5.4 Ammonia. 5.5 Ammonium acetate. chromatographically pure. 5.6 Ammonium acetate solution (5mmol/L). Weigh 0.38g of ammonium acetate (5.5), dissolve in water, and dilute to 1L. 5.7 Perchloric acid extract (0.4mol/L). Accurately measure 32.3mL of perchloric acid, and dilute to 1L with water. 5.8 Sodium hydroxide solution (2mol/L). Accurately weigh 8g of sodium hydroxide and dilute to 100mL with water. 5.9 Saturated sodium bicarbonate solution. Add enough solid sodium bicarbonate to water until it can no longer dissolve. 5.10 Hydrochloric acid solution (0.1mol/L). Accurately measure 8.3mL of concentrated hydrochloric acid, and dilute to 1L with water.
