4 Principle

The free solanesol (including the ester state) in potato leaves is extracted with n-hexane, and the total solanesol is saponified with sodium hydroxide to release the bound state. Solanesol, then remove impurities with water, centrifuge, take the upper solution diluted with mobile phase to volume, filter through a membrane and use high performance liquid chromatography- Determination by mass spectrometer, quantitative calculation by external standard method.

5 Reagents and materials

Unless otherwise specified, the reagents used are all analytically pure, and the water should meet the requirements of first-grade water in GB/T 6682. 5.1 n-hexane (C6H14). analytically pure. 5.2 Methanol (CH4O). chromatographically pure. 5.3 Isopropanol (C3H8O). chromatographically pure. 5.4 Sodium hydroxide (NaOH). analytically pure. 5.5 Absolute ethanol (C2H6O) is analytically pure. 5.6 Solanesol standard product (C45H74O, CAS number. 13190-97-1). purity ≥98%. 5.7 NaOH ethanol solution (0.02mol/L). Weigh 0.4g sodium hydroxide (5.4), dissolve it in absolute ethanol (5.5), and dilute to 500mL, stored in a polyethylene bottle, screw the cap tightly. 5.8 Mobile phase. methanol (5.2). isopropanol (5.3) = 50.50 (volume ratio), filtered through an organic phase filter membrane (5.11) before use, and ultrasonically degassed. 5.9 Solanesol standard stock solution (500μg/mL). Weigh 50mg solanesol standard product (5.6), accurate to 0.0001g, and place it in 100mL ......
