Foreword

Appendix A of this standard is a normative appendix. This standard is proposed and managed by the National Certification and Accreditation Administration. This standard was drafted. Beijing Entry-Exit Inspection and Quarantine Bureau of the People's Republic of China. The main drafters of this standard. Zhang Hexiao, Liu Huan, Liu Yanhua. This standard is the industry standard for inspection and quarantine issued for the first time. Marburg disease detection method

1 Scope

This standard specifies the method for detecting Marburg disease. This standard applies to the detection of antibodies to Marburg disease.

2 Abbreviations

The following abbreviations apply to this standard. 2.1 ELISA enzyme-linked immunosorbent assay. 2.2 IF immunofluorescence assay. 2.3 MBG Marburg disease. 2.4 PBS phosphate buffer. 2.5 TCID50 half the number of cell infections. 2.6 CPE cytopathic effect. 2.7 OD value optical density value.

3 Principle

3.1 IF test The cells infected with MBG virus are immobilized on a circular slide, and the antigen-antibody reaction is high in the presence of specific antibodies. The characteristic of degree specificity, together with the fluorochrome-labeled secondary antibody, binds to the antibody bound to the antigen and presents a fluorescence microscope. Specific fluorescence reaction. 3.2 ELISA test The antigen is coated on the ELISA plate, and in the presence of the specific antibody, the antigen and antibody are bound, and then the enzyme-labeled secondary antibody is added to form an antigen-antibody enzyme label. Secondary antibody complex. The enzyme in the complex catalyzes the decomposition of the substrate upon production of the corresponding substrate to produce a colored material. According to the presence or absence of colored substances And its concentration, indirectly speculate whether the antibody is present and its potency.
