Foreword

GB/T 20944 “Textiles - Evaluation for antibacterial activity” consists of three parts. - Part 1.Agar diffusion plate method; - Part 2.Absorption method; - Part 3.Shake flask method. This Part is Part 3 of GB/T 20944. This Standard was proposed by China Textile Industry Association. This Part shall be under the jurisdiction of Subcommittee on Basics of National Technical Committee on Textiles of Standardization Administration of China (SAC/TC 209/SC 1). 1 Scope

This Part of GB/T 20944 specifies quantitative test and evaluation method for determining the antibacterial properties of textiles by the shake flask method. This Part is applicable to various textile products such as down, fiber, yarn, fabric, and products with special shapes, especially applicable to non- dissolvable antibacterial textile products. This Part does not involve the evaluation of the safety of antibacterial products.

2 Normative references

The provisions in following documents become the provisions of this Part of GB/T 20944 through reference in this Standard. For dated references, the subsequent amendments (excluding corrigendum) or revisions do not apply to this Standard, however, parties who reach an agreement based on this Standard are encouraged to study if the latest versions of these documents are applicable. For undated references, the latest edition of the referenced document applies. 3 Terms and definitions

For the purposes of this document, the following terms and definitions apply. 3.1 antibacterial activity The activity of textiles to inhibit the growth and reproduction of bacteria on the fabric.

4 Safety precautions

Safety instructions. The test bacteria used in this test can cause human infection and disease.

5 Principle

Put the sample and the control fabric into an Erlenmeyer flask that is filled with a certain concentration of test bacteria liquid.

6 Test equipment

6.1 Spectrophotometer. The detection wavelength is 475nm or 660nm, suitable for testing the concentration of test bacteria. 6.2 Constant temperature incubator. temperature control accuracy is ±1°C. 6.3 Water bath. The temperature can be maintained at 46°C±2°C. 6.7 Autoclave. The temperature can be kept at 121°C and the pressure can be kept at 103kPa. 6.8 Erlenmeyer flask with stopper. The capacity is 250mL. 6.9 Petri dish. 90mm in diameter.

7 Medium and reagents

The reagents used in the test shall be analytically pure or suitable for microbiological tests. The test water shall be pure water, such as distilled water. 7.1 Nutrient broth 7.2 Nutrient agar medium 7.4 0.03mol/L PBS (phosphate) buffer Disodium phosphate 2.84g

8 Test strain

8.1 Strain Gram-positive bacteria. Staphylococcus aureus (ATCC 6538); Gram-negative bacteria. Escherichia coli (8099 or one of ATCC 11229, ATCC 8739, ATCC 29522), or Klebsiella pneumoniae (ATCC 4352). Choose one according to the needs; 8.2 Strain transformation and preservation After activation of the freeze-dried bacteria, inoculate the inclined tube culture. Then store in the refrigerator (5°C~10°C) as preservation bacteria. Planting shall be changed every month. Store in refrigerator (5°C~10°C) after planting. 8.3 Marks The following information shall be marked for each strain.

9 Preparation of test bacteria

9.1 Cultivation and preparation of bacterial liquid 9.1.1 Two-step pre-culture procedure to prepare bacterial inoculum suspension 9.1.2 Preparation of bacterial inoculum Use a pipette to draw 2mL~3mL from the bacterial suspension (9.1.1) (reference value. This step adjusts the number of live bacteria inoculated, the lower limit for Escherichia coli, and the upper limit for Staphylococcus aureus). 9.2 Cultivation and preparation of Candida albicans broth 9.2.1 Preparation of Candida albicans inoculum suspension by two-step pre-culture procedure Use a pipette to draw 2mL~4mL from the Candida albicans suspension (9.2.1). Transfer to a test tube containing 9mL of 0.03mol/L PBS buffer (7.4). Perform a 10-fold serial dilution operation. Mix well.

10 Preparation of sample

10.1 Sample washing If the sample needs to be washed, one of the 10.1.1 or 10.1.2 washing methods shall be used for operation. Indicate the washing method used in the test report. 10.2 Sample Cut the antibacterial fabric sample and the control fabric into pieces of about 5mm×5mm in size. Weigh 0.75g±0.05g as a sample. Wrap it in small pieces of paper. 10.3 Sample sterilization Put the small paper bag containing the sample into the autoclave. Sterilize at 121°C, 103kPa for 15min for later use.

11 Test operation

11.1 Sample and reagent bottling Prepare 9 250ml Erlenmeyer flasks. Respectively add 0.75g±0.05g of control fabric to each of the 3 flasks, and 0.75g±0.05g of antibacterial fabric sample to each of 3 flasks. The other 3 flasks are blank without sample. Then add 70mL±0.1mL of 0.03mol/L PBS buffer (7.4) to each flask. 11.3 "0" contact time sampling Use a pipette to draw 1mL ± 0.1mL of solution from each of the 6 flasks prepared with a contact time of "0". Transfer to a test tube containing 9mL ± 0.1mL of 0.03 mol/L PBS buffer (7.4). Mix well. Use 10 times dilution method to make another dilution. Mix well. Pipette 1mL±0.1mL into a sterilized petri dish, pour about 15mL of nutrient agar medium (7.2) or Sabouraud agar medium (7.3). 11.5 Dilution culture and determination of colony number After the specified time, draw 1mL±0.1mL of test solution from each flask. Transfer to a test tube containing 9mL±0.1mL of 0.03mol/L PBS buffer (7.4). Mix well. Use 10-fold dilution method to serially dilute to the appropriate dilution multiple. ......
