1 Scope

This standard specifies the use of high-performance liquid chromatography-diode array detector (HPLC-DAD) or high-performance liquid chromatography-tandem mass spectrometer. 3 Principle

Ultrasonic extraction of Michler's ketone and Michler's base in the sample with methanol. After the extract was filtered through a filter membrane, a high-performance liquid chromatography-diode array was used. Column detector (HPLC-DAD) or high performance liquid chromatography-tandem mass spectrometer (HPLC-MS/MS) measurement and confirmation.

4 Reagents and materials

4.1 Water. secondary water in accordance with GB/T 6682. 4.2 Methanol. chromatographically pure. 4.3 Mie's ketone reference material. CAS No. 90-94-8, purity ≥97.0%. 4.4 Mie base reference material. CAS No. 101-61-1, purity ≥98.0%. 4.5 Standard stock solution. Accurately weigh out the Michler's ketone reference material (4.3) and Michler's base reference material (4.4), dissolve and make up the volume with methanol (4.2) Prepare a standard stock solution with a concentration of 1000 mg/L, respectively. 4.6 Mix standard working solution. Accurately draw the appropriate amount of standard stock solution (4.5), and dilute with methanol (4.2) to at least 5 different concentrations. Series mix standard working solution. When using HPLC-DAD, the concentration can be formulated as 0.5mg/L, 5mg/L, 10mg/L, 15mg/L, 20mg/L working solution; when using HPLC-MS/MS, it can be prepared at a concentration of 20μg/L, 40μg/L, 80μg/L, 160μg/L, 200 μg/L working solution. Note. The standard stock solution and the mixed standard working solution are stored in a refrigerator at 0 ° C to 4 ° C, protected from light. The validity period of the standard stock solution is 6 months. The solution is valid for 1 month.
