Amphibian acute toxicity test1 ScopeThis Part of GB/T 31270 specifies the amphibian acute toxicity test’s material, conditions, operation, quality control, data processing, test report and other basic requirements. This Part is applicable to amphibian acute toxicity test for chemical pesticide registration. Other types of pesticides may refer for use. This Part is not applicable to volatile and insoluble chemical pesticides.2 Terms and definitionsThe following terms and definitions are applicable to this document. 2.1 Median lethal concentration In the acute toxicity test, the concentration of test substance when it causes 50% of test biology dead, represented in LC50. Note. The unit is mg a.i./L. 2.2 Test substance The substance to be tested in the test. 2.3 Chemical pesticide The pesticide that is synthetically made of chemical substances. Some take active substance in natural product as parent body to imitate and structurally transform, which are called biomimetic synthesis pesticides. Synonym. synthetic organic pesticide [NY/T 1667.1-2008, Definition 2.3.1] 2.4 Technical material The final product that is made of active ingredients and impurities obtained in the manufacturing process. It must not contain visible foreign matters or any additives. If necessary, add a small amount of stabilizer. [NY/T 1667.2-2008, Definition 2.5.1] 2.5 Formulation product The product that is made of pesticide (or parent drug) and additive, and has stable state of use. [NY/T 1667.2-2008, Definition 2.4] 2.6 Semi-static test During the test, replace the liquid at a regular interval (e.g. 24h), so as to maintain the concentration of the test liquid not less than 80% of the initial concentration. 2.7 Static test Do not replace the test liquid during the test. 2.8 Flow-through test Continuously refresh the test liquid during the test.3 Test overviewThe test methods for amphibian acute toxicity are static test, semi-static test, and flow-through test. It shall adopt suitable method according to the nature of the test substance. Respectively prepare different concentrations of the test substances. Observe and record tadpole’s symptoms of poisoning and number of deaths every day during 96 h test. And calculate LC50 values and 95% confidence limits of 24 h, 48 h, 72 h and 96 h respectively.4 Test methods4.1 Material and conditions 4.1.1 Test biology Rana Limnocharis or Xenopus laevis tadpole is recommended. Specific age and appropriate water temperature can refer to Appendix A. If use other amphibians as test material, they shall meet corresponding domestication and test conditions of physiological requirements, and explanation shall be given as - Mortality rate during the test shall not exceed 10%, and no abnormal behavior occurs; - During the test, the dissolved oxygen content of test liquid shall not be less than 60% of air saturation value (ASV).5 Test reportTest report shall at least contain the following items. - Information of test substance, including test pesticide’s common name, chemical name, structural formula, CAS number, purity, basic physical and chemical properties, origin, etc.; - Test biology’s name, origin, size and domesticated situation; - Test conditions, including test temperature, illumination, dilution water’s temperature, dissolved oxygen concentration and pH, etc.; - Test liquid concentration, LC50 value of 24 h, 48 h, 72 h, and 96 h, 95% confidence limits, and calculation method; - If tadpoles of the control group die or have abnormal reactions; - Observed toxic effects, e.g., any abnormal behavior or poisoning symptom of test tadpoles; - Classification of amphibian toxicity refers to Appendix B.Appendix B(Informative) Acute toxicity classification of pesticide to tadpole According to median lethal concentration LC50 (96h) of pesticide to tadpole, classify the acute toxicity of pesticide to tadpole into four grades, as shown in Table B.1.
