Introduction

Virus, IBV) is an acute, highly contagious infectious disease of chickens. IB can occur in chickens of various ages, chickens less than 5 weeks old Especially susceptible, and the symptoms are more obvious. The clinical features of IB are mainly manifested as dyspnea, rales, coughing and sneezing; decreased egg production. Low, egg quality declines; chicks appear to die. IB is widely prevalent all over the world and seriously endangers the production of chicken industry. IBV belongs to Coronaviridae, Coronaviridae, and Gammavirus in classification, and is a single-stranded positive-sense RNA virus whose RNA is easily In terms of variation, IBV has diversity. Most IBV strains can cause specific lesions in the trachea, but some strains can cause kidney lesions and Genital tract lesions. At present, the IBV strains that have been isolated at home and abroad belong to more than 30 serotypes, and the cross-immunity between IBV strains of each serotype Low protection. The diagnosis of IB is mainly carried out by etiological and serological tests. Due to the widespread use of IBV live strain vaccines in production to prevent this disease, this is Clinically, it increases the difficulty of diagnosing IB. This document standardizes the clinical diagnosis technology of IB; standardizes the pathogenic diagnosis technology of IB, including virus isolation and identification, virus RT- PCR, virus real-time fluorescent RT-PCR, virus genotype identification; standardized IB serological diagnostic techniques, including hemagglutination and hemagglutination inhibition tests Test, tracheal ring tissue culture serum neutralization test. These diagnostic techniques are also recommended in the OIE Manual of Terrestrial Animal Diagnostic Tests and Vaccines. In this revision process, references were made to the relevant diagnostic techniques recommended in this manual. Diagnosis Technique of Chicken Infectious Bronchitis

1 Scope

This document specifies the clinical diagnosis, virus isolation and identification, virus RT-PCR, and virus real-time fluorescence of chicken infectious bronchitis (IB). Technical requirements for RT-PCR, virus genotype identification, hemagglutination and hemagglutination inhibition test, tracheal ring tissue culture serum neutralization test. This document is applicable to the diagnosis, quarantine, surveillance and epidemiological investigation of IB.

3 Terms and Definitions

The following terms and definitions apply to this document. 3.1 chicken infectious bronchitis Avian infectious bronchitis virus that mainly infects chickens caused by avian infectious bronchitis virus belonging to the Coronaviridae, Coronaviridae, Gammavirus genus disease. 3.2 hemagglutinin The process of agglutinating chicken erythrocytes with the antigen obtained after the culture solution of the IBV strain is digested with trypsin. Note. By selecting appropriate IBV strains and preparing IBV hemagglutination antigens, the serological diagnosis of IB is carried out. 3.3 The IB antibody specifically attaches to the specific site of the IBV hemagglutination antigen and interferes with the binding between the IBV hemagglutination antigen and the red blood cell receptor. Together, inhibit the ability of IBV hemagglutination antigen to agglutinate chicken red blood cells. 3.4 Ct value cyclethreshold The number of cycles it takes for the fluorescent signal amount in each real-time fluorescent RT-PCR reaction tube to reach the set threshold.
