Animal-derived protein feed pepsin Determination of digestibility by filtration method

1 Scope

This document describes a method for determining the pepsin digestibility of protein feedstuffs of animal origin. This document applies to the determination of pepsin digestibility of animal-derived protein feeds.

2 Normative references

The contents of the following documents constitute the essential clauses of this document through normative references in this document. For referenced documents without a date, only the version corresponding to that date applies to this document; for referenced documents without a date, the latest version (including all amendments) applies to This document. 3 Terms and Definitions

The following terms and definitions apply to this document. 3.1 At 45℃ and pH 1-2, animal-derived protein feed was digested with 20U/mL pepsin solution for 16h, and the digested crude protein The ratio of protein to crude protein. Note. There is no direct correlation between pepsin digestibility and in vivo digestibility of animal-derived protein feeds.

4 Principles

The sample was digested with pepsin under specified conditions, the insoluble residue was separated, and the undigested crude protein content in the residue and the crude protein content of the sample were determined. Protein content was calculated and pepsin digestibility was obtained.

5 Reagents or materials

Unless otherwise specified, only analytical grade reagents were used. 5.1 Water. GB/T 6682, Grade 3. 5.2 Petroleum ether. boiling range 30℃~60℃. 5.3 Anhydrous ethanol. 5.4 Pepsin. Derived from porcine stomach, CAS No. 9001-75-6.Enzyme activity is determined according to Appendix A. 5.5 Hydrochloric acid solution. Pipette 6.1 mL of hydrochloric acid, dilute to 1000 mL (pH 1-2) with water, and mix thoroughly.
