General rules
1 Sample collection and precautions
1.1 The collected samples shall be representative. Generally, the corresponding number
of packaging units are randomly selected depending on the size of each batch of
cosmetics. During the inspection, totally 10 g or 10 ml shall be taken from samples
from more than two packaging units. The sample size can be reduced as appropriate for
samples with small packaging volume.
1.2 The samples for inspection shall strictly maintain the original packaging state. The
container shall not be broken and shall not be opened before inspection, to prevent re-
contamination.
1.3 After receiving the sample, it shall be registered and numbered immediately; the
inspection shall be carried out as soon as possible according to the inspection
requirements. If it cannot be inspected in time, the sample shall be placed in a cool and
dry place at room temperature; it shall not be refrigerated or frozen.
1.4 If there is only one sample and multiple analyses are required at the same time, such
as bacteria, toxicology, chemistry, etc., it should take out part of the sample for bacteria
inspection first; then the remaining sample is used for other analyses.
1.5 During the inspection process, from opening to the end of all inspection operations,
re-contamination and spread of microorganisms must be prevented. All utensils and
materials used shall be sterilized in advance; all operations shall be carried out in a
sterile room. Or under appropriate conditions, it is carried out according to the aseptic
operation requirements.
1.6 If fecal coliform bacteria or other pathogenic bacteria are detected, the strains and
samples shall be kept for one month from the date of the report, for future reference.
2 Preparation of samples for inspection
2.1 Culture medium and reagents
2.1.1 Physiological saline

Sodium chloride. 8.5 g
Distilled water. 1000 ml
After dissolution, dispense it into conical bottles with glass beads, 90 ml per bottle, to
perform high pressure sterilization at 121 °C (15 lb) for 20 min.
2.1.2 SCDLP liquid culture medium
Ingredients. Casein peptone. 17 g
Soy peptone. 3 g
Sodium chloride. 5 g
Potassium hydrogen phosphate. 2.5 g
Glucose. 2.5 g
Lecithin. 1 g
Tween 80.7 g
Distilled water. 1000 ml
Preparation method. Mix the above ingredients. Heat to dissolve. Adjust pH to 7.2 ~
7.3.Sterilize it at high pressure at 121 °C (15 1b) for 20 min. Pay attention to oscillate
to fully mix the Tween 80 precipitated at the bottom. Cool to about 25 °C before use.
Note. If casein peptone and soybean peptone are not available, Japanese polypeptone can also
be used instead.
2.1.3 Sterilized liquid paraffin.
2.1.4 Sterilized Tween 80.
2.2 Instruments
2.2.1 Balance.
2.2.2 Sterilized conical flask. Contains glass beads and 90 ml diluent.
2.2.3 Sterile graduated pipette. 10 ml, 5 ml.
2.2.4 Water bath.
2.2.5 Sterile mortar and pestle.
2.2.6 Homogenizer.
