Determination of crude protein in feeds - Kjeldahl method
1 Scope
This standard specifies the Kjeldahl method for the determination of crude protein in feed.
This standard is applicable to the determination of crude protein in feed ingredients, compound feed, concentrated feed, concentrate supplement and additive premix feed.

3 Principle
The sample is digested by sulfuric acid under the action of a catalyst, and the nitrogen-containing compound is converted into ammonium sulfate, and alkali is distilled to make the ammonia escape, and after absorption with boric acid,
Then titrate with a standard hydrochloric acid titration solution, measure the nitrogen content, multiply by 6.25, and calculate the crude protein content.
4 reagents or materials
Analytically pure reagents were used unless otherwise stated.
4.1 Water. GB/T 6682, Level 3.
4.2 Boric acid. chemically pure.
4.3 Sodium hydroxide. chemically pure.
4.4 Sulfuric acid. chemically pure.
4.5 Ammonium sulfate.
4.6 Sucrose.
4.7 Mixed catalyst. Weigh 0.4g of copper sulfate pentahydrate, 6.0g of potassium sulfate or sodium sulfate, grind and mix; or purchase commercial Kjeldahl
Catalyst sheet.
4.8 Boric Acid Absorbent I. Weigh 20 g of boric acid, dissolve in water and dilute to 1000 mL.
4.9 Boric Acid Absorbent II. 1000 mL of 1% boric acid aqueous solution, 10 mL of 0.1% bromocresol green ethanol solution, 0.1% methyl red ethanol
7 mL of the solution, 0.5 mL of a 4% aqueous sodium hydroxide solution, and mix well, and the storage period at room temperature is 1 month (for automatic procedures).
4.10 Sodium hydroxide solution. Weigh 40g of sodium hydroxide, dissolve it in water, and after cooling to room temperature, dilute to 100mL with water.
4.11 Hydrochloric acid standard titration solution. c (HCl) = 0.1mol/L or 0.02mol/L, prepared and calibrated according to GB/T 601.
4.12 Methyl red ethanol solution. Weigh 0.1 g of methyl red, dissolve it with ethanol and dilute to 100 mL.
4.13 Bromocresol green ethanol solution. Weigh 0.5g of bromocresol green, dissolve it with ethanol and dilute to 100mL.
4.14 Mixed indicator solution. Mix the methyl red ethanol solution and the bromocresol green ethanol solution in equal volumes. The solution is stored at room temperature and protected from light.
