3 Terms and definitions

4 Principle
The sample is extracted with water to remove impurities such as fat, protein, starch, non-starch sugar, and the reducing end of xylobiose is combined with 1-phenyl-3-methyl-5-pyrazole
The derivatization reaction of PMP occurs to produce a stable product with strong ultraviolet absorption, which is detected by high-performance liquid hydrophilic retention chromatography.
Measured and quantified by external standard method.
5 Reagents and materials
Unless otherwise specified, the reagents used in this method are all analytically pure, and the water is the first grade water specified in GB/T 6682.
5.1 Reagents
5.1.1 Acetonitrile (CH3CN). chromatographically pure.
5.1.2 Anhydrous ethanol (CH3CH2OH).
5.1.3 Acetone (CH3COCH3).
5.1.4 Methanol (CH3OH).
5.1.5 1-Phenyl-3-methyl-5-pyrazolone (PMP) (C10H10N2O).
5.1.6 Trichloromethane (CHCl3).
5.1.7 Hydrochloric acid (HCl).
5.1.8 Sodium hydroxide (NaOH).
5.1.9 Phosphoric acid (H3PO4). chromatographically pure.
5.1.10 Xylobiose (C10H18O9, CAS number. 6860-47-5). Purity ≥99.0%.
