1 Scope
This standard specifies the qualitative and quantitative HPLC method for the content of astragaloside, paeoniflorin, forsythin and forsythiaside A in cosmetics.
The principle, equipment and materials of the detection method, analysis steps, calculation of results, recovery and precision, allowable difference, etc.
This standard applies to the determination of the content of astragaloside, paeoniflorin, forsythin and forsythiaside A in water, emulsion and cream.
The detection limit of this standard method for astragaloside IV is 15mg/kg, the limit of quantification is 45mg/kg; the detection limit of paeoniflorin is 3mg/kg,
The limit is 8 mg/kg; the detection limit of forsythin and forsythiaside A is 20 mg/kg, and the limit of quantification is 60 mg/kg.

3 astragaloside IV
3.1 Principle
Astragaloside is dissolved in hot water and then extracted with water-saturated n-butanol, washed with ammonia solution, purified by C18 solid phase extraction column, high performance liquid chromatography
Separation, evaporative light scattering detector measurement, external standard method for quantification.
3.2 Reagents and materials
Unless otherwise specified, the reagents used are chromatographically pure, and the water is the first grade water specified in GB/T 6682.
3.2.1 Methanol.
3.2.2 20% methanol solution. Measure methanol (3.2.1) 20 mL and mix with 80 mL of water.
3.2.3 n-butanol. analytically pure.
3.2.4 Acetonitrile.
3.2.5 Ammonia.
3.2.6 Ammonia test solution. Take 400 mL of ammonia water (3.2.5) and dilute to 1 L with deionized water.
3.2.7 Astragaloside standard substance. purity is not less than 96%. Chinese and English name, CAS number, molecular formula and relative molecular mass of astragaloside

3.2.8 Standard working solution of astragaloside. accurately weigh 100mg of astragaloside standard substance (3.2.7) (accurate to 0.1mg)
In a 100mL volumetric flask, dilute to the mark with methanol, prepare a standard stock solution of astragaloside IV at a concentration of 1000mg/L, and avoid it at 4°C.
Light preservation, valid for 2 months. Accurately transfer 2 mL, 3 mL, 4 mL, 5 mL, 10 mL, 50 mL of the standard stock solution of Astragaloside
After 100 mL of the brown volumetric flask, the volume was adjusted to the mark with methanol, and the mass concentrations were 20 mg/L, 30 mg/L, and 40 mg/L, respectively.
50mg/L, 100mg/L, 500mg/L of the standard working solution of astragaloside, stored at 4 ° C in the dark, can be used for one week.
3.2.9 C18 solid phase extraction column..200 mg, 3 mL.
