4 reagents and materials
Unless otherwise stated, the reagents used were of analytical grade and water was a primary water specified in GB/T 6682.
4.1 methanol. pure chromatography.
4.2 heptafluorobutyric acid. pure chromatography.
4.3 alkaline phosphatase. enzyme activity of not less than 10U/mg.
4.4 phosphoric acid. excellent grade pure.
4.5 ammonia. excellent grade pure, content of 25% to 28%.
4.6 ammonium dihydrogen phosphate. excellent grade pure.
4.7 Disodium hydrogen phosphate (Na2HPO4 · 2H2O).
4.8 Ethanol.
4.9 Hydrochloric acid.
4.10 aqueous solution of potassium hydroxide. Weigh 250g of solid potassium hydroxide and add.200 mL of water to dissolve.
4.11 30% phosphoric acid aqueous solution (volume fraction). Accurately measure 30mL phosphoric acid (4.4) and 70mL water, mix and spare.
4.12 30% ammonia solution (volume fraction). Accurately measure 30mL ammonia (4.5) and 70mL water, mix and spare.
