GB/T 29371 Technical rules for seed producing system of two-line hybrid rice consists of
five parts, as follows.
— Part 1. Terminology;
— Part 2. Technical rules for foundation seed production of photo/thermo-sensitive genic
male sterile line;
— Part 3. Technical rules for seed multiplication of photo/thermo-sensitive genic male
sterile line;
— Part 4. Technical rules for F1 seed production;
— Part 5. Technical rules for identification of variety purity & male sterility of MS line.
This is Part 2 of GB/T 29371.
This Part is drafted according to the rules specified in GB/T 1.1-2009.
This Part was proposed by Ministry of Agriculture of the People’s Republic of China.
This Part shall be under the jurisdiction of National Technical Committee (SAC/TC 37) on
Crop Seed of Standardization Administration of China.

sterile line
1 Scope
This Part of GB/T 29371 specifies the methods and procedures to produce the high
temperature inducing male rice photo/thermo-sensitive genic male sterile line foundation
seed, in the phytotron (box) and cold water processing pond.
This Part applies to the high temperature inducing male rice photo/thermo-sensitive genic
male sterile line foundation seed production.
2 Facilities construction
2.1 Artificial climate box
2.1.1 The volume of the artificial climate box
Artificial climate box’s interior area is not less than 3m2 and its height is not lower than 2m.
2.1.2 Parameters settings of light, temperature and humidity of the artificial
climate box
It shall comply with provisions indicated in Table 1 and Table 2.
When the test plants grow to 5 to 6 leaves, transplant them to some plastic buckets with a
diameter of appropriately 28cm. Each bucket contains 4 plants. They grow under natural
temperature condition and under normal management after transplanting. When the
stems grow to the early-stage of the young-ear differentiation stage IV (indica rice), or
stage II (japonica rice), select those single-plants which are in uniform growth status.
Place into the artificial climate box. Setup the light and temperature parameters in
accordance with the standard of male sterile line identification (examination). Transplant
them to a natural condition when the stems and ears grow up to the end-stage of stage VI,
or the flag-leaf’s pulvinus distance is 0cm-2cm. After the treatment, the single-stem with a
flag-leaf’s pulvinus distance of 0cm-2cm shall be individually tagged, numbered and
recorded. After the tagged stems are eared, microscope-inspect the pollen and bag them;
and inspect the pollen fertility. After the maturity, examine the selfing setting rate. Select
the single-plant, of which both of the pollen fertility and the spikelet fertility are zero, as the
core single-plant.
2.2 Phytotron
2.2.1 Structure
The usage area of the single phytotron is 12m2 - 15m2, and the height is 4m - 4.5m. The
thickness of the wall is 30m - 40m. It is equipped with the thermal insulation material
inside the wall. There is transparent thermal insulation layer at the place 2.0m above the
ground. Adopt the 8mm tempered glass. Above the insulation layer, install artificial light
source. At the southern-wall, set up a ventilating window with the size of 1m X 1.5m, at
0.5m above the ground. Ventilating window shall adopts the double glazing with the
thickness of 5mm - 8mm. It needs to set up a good ventilation system in the phytotron.
And it is equipped with the water supply and water storage device.
2.2.2 Scale
The control system adopts automatic control equipment. And the scale of the control
chamber shall be set up according to the number of the phytotrons.
2.2.3 Temperature control equipment
Four sets of wall-mounted air conditioning with the refrigeration and heating capacity of
3kw - 4kw are installed inside the phytotron, one set on every wall, dislocation-installed
under the glass thermal insulation layer. Temperature control indicators of the temperature
control equipment shall be set within (15°C - 30°C)±1°C.
2.2.4 Artificial light source
The indicator of indoor light intensity is (1.0-2.0) X 104 lx. The artificial light source shall
adopt the 400W high pressure sodium lamp, in combination with the 400W metal halide
lamp. The Ratio is 3.1. The quantity of light source lamps is 15-20, with electrical
automatic light-adjustment rectifier.

2.3.3 Temperature control
Set 4 water inlets and outlets respectively. Place the mercury thermometers at the
sensitive parts of fertility transform temperature of sterile line of inlets and outlets.
2.4 Natural cold water pond
Use the low-temperature water in reservoir or cold underground water, with the
temperature of water source of ≤20°C. Depth of cold water pond is ≥40cm, with an area of
cold water pond of 30m2 - 60m2. The flow rate of cool water is ≥ 6m3/h - 12m3/h. A mercury
thermometer shall be placed at the position of inlet and outlet.
3 Source of foundation seed
For the sterile line that is newly approved or identified, use the breeder seeds as the seed
source. For sterile line that has been produced and applied extensively, select the core
single-plant from the foundation seed as the seed source.
4 Production of core seed
4.1 Preparation of cultivation pot
Select the plastic pot with a body-height of 20cm-25cm and a diameter of 25 cm-30 cm.
The loading amount of mud shall be appropriate that the mud level is 5cm away from the
port edge. The ordinary field soil shall be used.
4.2 Nursery for single-plant
Conduct the seed soaking, germination acceleration, and seedling according to
conventional method. When it grows to 5-leaves 1-heart, transplant them into the field, the
density is 20 cm X 20 cm, transplant more than 1000 plants of single-plant. Cultivate and
manage it according to general field production.
4.3 Transplant
When the stem of single-plant selection nursery plants come into the inflorescence
differentiation stage II (japonica rice) or stage 1 (indica rice), select those plants with
uniform growth and typical properties of the male sterile line. Transplant into the prepared
cultivation pots with soil, on a cloudy day or at the evening time. 4 plants in one pot, or
directly transplant them into the cold water pond with soil. Ensure adequate water supply
and prevent from insect disease.
4.4 Low temperature processing
4.4.1 Processing in phytotron

5.1 Multiplication methods
Multiplication by cold water irrigation, multiplication in autumn, or multiplication in winter.
5.2 Cultivation and management
The seeds of core single-plant collected are divided into single-plant soaking seed,
accelerating germination, sowing, and individual-plant planting. Each single-plant is
planted in one plant-line. Field cultivation and management can refer to the multiplication
technology of photo/thermo-sensitive genic male sterile line.
5.3 Observation and identification
Respectively observe and identity the plant-lines at tillering period, heading and flowering
period, and maturation period. At anytime, eliminate the plant-lines with character
variation and segregation.
5.4 Harvesting
Mix-harvest the seeds of selected plant-lines, that is the foundation seed.
