1 Scope

This standard specifies the method for the determination of amino acids in chemical fibers with an automatic amino acid analyzer. This standard applies to aspartic acid, threonine, serine, glutamic acid, proline, glycine, alanine, cystine, and valine in chemical fibers. 3 Principle

The protein in chemical fiber is hydrolyzed into free amino acid by hydrochloric acid, and separated by the ion exchange separation column of the amino acid analyzer, The triketone solution produces a derivative color reaction, and then the amino acid content is determined by colorimetry and quantified by external standard method.

4 Reagents and materials

4.1 Reagents 4.1.1 Concentrated hydrochloric acid, excellent grade pure. 4.1.2 Water, in line with the first grade water specified in GB/T 6682. 4.1.3 Sodium citrate (Na3C6H5O7·2H2O), excellent grade pure. 4.1.4 Sodium hydroxide (NaOH), excellent grade pure. 4.1.5 Sodium chloride (NaCl), excellent grade pure. 4.1.6 Lithium hydroxide (LiOH·H2O), excellent grade pure. 4.1.7 Glacial acetic acid (C2H4O2), excellent grade pure. 4.1.8 Dimethyl sulfoxide (C2H6OS), pure superior grade. 4.2 Reagent preparation 4.2.1 6mol/L hydrochloric acid. Mix concentrated hydrochloric acid and water at a ratio of 1.1 (volume ratio). 4.2.2 0.02mol/L hydrochloric acid.
