categories of pathogenic microorganisms in the Catalogue of Pathogenic Microorganisms
Transmissible to Humans.
The various culture medium components used to culture bacterial species shall comply with the
requirements of the bacterial species preservation and management institutions.
A.2 Reagents and Equipment
A.2.1 Reagents
A.2.1.1 Nutrient agar (NA) medium
Beef extract 5.0 g
Peptone 10.0 g
Sodium chloride 5.0 g
Agar 15.0 g
Preparation method. dissolve the other components except agar in 1, 000 mL of distilled water,
use 0.1 mol/L NaOH solution to adjust the pH to 7.2 ~ 7.4, add agar, heat to dissolve it, divide
the contents into portions, and at 121 C, sterilize them in a pressure steam sterilizer for 20 min.
The above formula is prepared as nutrient broth medium without adding agar.
If commercial culture medium is used, it shall be prepared and sterilized in accordance with the
preparation method and sterilization conditions specified on the culture medium.
A.2.1.2 Tryptone soy broth (TSB) medium
The concentration of the bacterial suspension can also be roughly estimated using turbidimetry,
that is, measuring the absorbance of the bacterial suspension at a wavelength of 600 nm.
If the prepared bacterial suspension is not immediately used, it shall be stored at 4 C and used
within 2 hours.
A.5 Test Steps
A.5.1 Preparation of bacterial slices
The bacterial slices are made by dripping bacterial solution onto a carrier, and the carrier is a
water-based microporous filter membrane (pore size 0.45 m, diameter 50 mm 5 mm). During
bacteria contamination, spread the sterilized dry carrier slice flat on a sterile plate and dropwise
add bacterial solution to each slice, with (200 ~ 300) L per slice. The number of recovered
bacteria on each bacterial slice, as obtained by counting the viable bacteria culture, shall be no
less than 106 CFU/slice. The bacterial slices shall be dried naturally at room temperature before
use.
Before testing, keep the bacterial filter membrane in a fully moistened state (if a small amount
of bacterial solution cannot fully moisten the filter membrane, add sterile water to moisten the
filter membrane first).
If the test site has a complete flat surface, the bacterial solution can also be directly applied to
the corresponding position.
A.5.2 Placement of bacterial slices
Evenly place the plate with moist bacterial slices in the gap inside the indoor unit body and
close the wind deflector.
In order to ensure that the plate can be stably placed in the prototype, compared to the actual
installation state, the installation state and some structures of the test prototype may need to be
changed, for example, maintaining the wall-mounted air conditioner in an inclined position.
The bacterial slices can also be pasted on the surface of each component and part that needs to
be tested.
A.5.3 Self-cleaning test
Start the program corresponding to the self-cleaning function of the test prototype. The test time
shall comply with the time required by the appliance, but shall not exceed 4 hours. Evenly
arrange the bacterial slices on the filter screen, evaporator, fan, the inner rear part of the rear
frame shell, and the middle positions of the upper and lower air guide blades, and the left and
right air guide blades of the prototype, with 2 slices evenly arranged in each position, for a total
of 12 slices.
If necessary, sampling at other locations may also be added.
